A peptide purity result and an endotoxin result answer different questions. A clean-looking chromatogram does not establish that a research sample is low in bacterial endotoxin. To assess that, look for a separate test, its reporting units and evidence that the sample did not interfere with the method.12

What endotoxins are

Endotoxins are lipopolysaccharides, usually shortened to LPS, associated with the outer membrane of Gram-negative bacteria. They are bacterial material, not a peptide synthesis by-product. Endotoxin can remain after bacteria are no longer viable, so a sterility result and an endotoxin result are not interchangeable.3

Synthetic origin does not make this question irrelevant. An experimental study of reagents used in microglia research detected LPS in several preparations, including synthetic peptides. Its functional experiments showed that contaminating LPS could account for some apparent inflammatory activity. That finding is a reason to check the actual material, not to assume that every synthetic peptide is contaminated.4

Why it matters in cell and animal research

If an experiment measures inflammatory signalling, an unmeasured inflammatory stimulus is a confounder. The microglia study examined changes in cell markers and mediator release; some responses diminished when LPS activity was blocked. A response attributed to the named reagent can therefore need a second explanation.4

The same distinction matters when interpreting animal research: endotoxin has biological effects of its own, including inflammatory responses. The acceptable background for a particular model must be justified for that experiment. A product specification is not automatically a no-effect threshold for every cell system or animal model.34

How the LAL test works

LAL stands for Limulus amebocyte lysate, a reagent made from horseshoe-crab blood cells. Endotoxin activates an enzyme cascade in the reagent. Depending on the method, the laboratory detects a clot, a change in turbidity or a colour-producing reaction. These are controlled analytical reactions; cloudiness in the original peptide vial is not an LAL result.5

Method suitability matters. Sample ingredients can suppress or enhance the response, so a result needs appropriate controls and interference checks. Ask whether the laboratory checked recovery of added endotoxin in the sample matrix, and what dilution and reporting limit were used. A bare “pass” leaves those questions unanswered.25

LAL is not the only available approach. FDA's 2026 update also recognises the broader use of recombinant reagents, with suitability established for the material being tested. A report should identify its actual method rather than treating every endotoxin assay as LAL.2

What EU/mg means

EU means endotoxin units: activity measured relative to a reference standard. EU/mg expresses that activity per milligram of material. It is neither a percentage purity nor a count of living bacteria. EU/mL and EU/vial use different denominators, so copy the unit as carefully as the result.5

A hypothetical example makes the distinction clear: if a report gives 2 EU for a vial containing 10 mg of measured peptide, dividing 2 by 10 gives 0.2 EU/mg of peptide. If the denominator were gross powder mass instead, the description would need to say so. These are illustrative numbers, not a Moleano result or an acceptance limit.

Keep inequality signs too. “Less than 0.1” is a bound under the stated test conditions, not a measured zero. When comparing documents, first align the units, mass basis and reporting limits; otherwise the smaller-looking number may not represent the lower endotoxin burden.25

Does a cloudy peptide solution indicate endotoxin?

Appearance cannot establish endotoxin content. Peptide solubility depends on sequence and solvent conditions, and incomplete dissolution can explain visible material. Follow the peptide's data sheet and the intended assay requirements when investigating a cloudy peptide solution; do not diagnose it from a photograph. Our solubility troubleshooting guide explains the separate dissolution question.6

A clear solution still needs the appropriate analytical evidence. HPLC main-peak area percentage describes detected chromatographic components; it is not a substitute for an endotoxin assay. Likewise, an endotoxin report does not establish peptide identity or content.15

Which Moleano reports are published?

The Lab Reports page publishes endotoxin (LAL) reports for 13 catalog entries: AOD-9604, BPC-157, NAD+, TB-500, MP-3 RETA (retatrutide), tesamorelin, CJC-1295 without DAC, the BPC-157/TB-500 blend, MOTS-c, sermorelin, GHK-Cu, MP-1 SEMA (semaglutide) and MP-2 TIRZ (tirzepatide). This list describes the published report inventory, including the blend; it is not a claim that every current lot has the same result.7

Open the report itself and compare the sample identifier, date, method and units with the material under consideration. If a report uses EU/vial, retain that unit unless a justified conversion is available. For a practical starting point, see BPC-157 and its published documents on Lab Reports.

Frequently asked questions

Does high peptide purity establish low endotoxin?

No. Chromatographic purity and endotoxin are separate measurements. Look for an endotoxin result with its method, units, reporting limit and sample identifier.

What does EU/mg mean?

Endotoxin units per milligram of the stated material. Check whether the denominator is peptide mass or gross material; EU/mg, EU/mL and EU/vial are not interchangeable.

Does a cloudy peptide solution prove endotoxin contamination?

No. Appearance cannot establish endotoxin content. Investigate dissolution against the data sheet and assess endotoxin using an appropriate analytical test.
